Sprang Cell, Vol 89, 251-261, April 19, 1997 This kinemage contains the RGS 4 Guanine Tri-phosphate Activating protein (GAP) complexed with one of its physiological substrates, the Gi-Alpha 1 subunit "loaded" with GDP-AlF4-Mg(2+). The metal/ dinucleotide component of the crystal simulates the transition state. Both proteins are biologically active as monomers and were cloned from the brain of the RAT. The crystallized protein structure was made with purified RGS4 and Gi Alpha 1 purified from E. coli. coordinates from file 1AGR.pdb The RGS family of proteins (Regulators of G protein Signaling) function to increase the rate of GTP hydrolysis by heterotrimeric G proteins. It has been appreciated for some time that there exists complex signaling cascades within Eukaryotic cells which couple extracellular stimuli with intracellular events. Heterotrimeric G proteins consist of a an Alpha subunit and a Beta-Gama complex. The Alpha subunit is bound to GDP in its inactive state, and associated with the beta-gamma complex. Upon signaling (through a receptor for instance) the G alpha subunit exchanges GDP for GTP and becomes active. The GTP bound Alpha subunit disassociates from the beta-gamma complex and activates its targets: ion channels, nucleotide phosphodiesterases, adenylate cyclase and phospholipases. The signaling stops when the intrinsic GTPase ability of the G Alpha subunit hydrolyzes GTP to GDP. Hence, while in the GTP bound state, G Alpha proteins are able to activate their effectors. The catalytic rate of intrinsic GTP hydrolysis is slow, often on the order of minutes. The RGS family increases the intrinsic rate of GTP hydrolysis substantially. They increase the intrinsic catalytic activity of GTP-Alpha, in effect inactivating it. Thus RGS proteins serve as an important signal regulation unit and no doubt will be implicated in different diseases that involve hyperactive G Alpha subunits. An important question to ask is, "how does the RGS family increase GTP hydrolysis?" They accomplish this by stabilizing the transition state of GTP-bound G Alpha. Experiments using different analogs of GTP bound Gi Alpha1, which forces the protein to mimic different conformations, show that RGS4 has higher affinity for the transition state than for the GTP or GDP bound forms. Catalytic studies have determined that RGS4 acts by increasing hydrolysis rather than slowing GDP release. The molecular interactions as suggested by site directed mutagenesis and the data from various crystal structures, suggest that RGS4 acts by stabilizing the transition state and not by introducing sidechains into the active site. This mechanism is distinct from GAPs (GTPase Activating Proteins) among the small G Protein family (such as suggested for p120 gap) which may function by coordinating catalytic water molecules, Mg2+, or stabilizing the transition state GTP molecule. It will be interesting to see if other members of the RGS family function through stabilization of the switches, or if they use sidechains to coordinate other sidechains, water, Mg2+ or the transition state GTP molecule. Kinemage 1 shows the overall crystal structure and highlights the "Switch" regions of the Gi Alpha 1 protein. RGS 4 stabilizes the three switch residues, providing rigidity to the molecule and it is this interaction that is thought to increase GTP hydrolysis. Kinemage 2 shows the RGS fold and the conserved regions among the RGS family. Kinemage 3 shows the molecular interactions between the Switch regions of Gi Alpha 1 and the highly conserved RGS Fold. Notice that the RGS 4 protein contains nine alpha helices that form the family conserved RGS domain. The crystal structure unfortunately only includes residues 5 to 354 for Gi Alpha 1 and 51 to 178 for RGS4. The unit cell of the crystal lattice contained two RGS4/ Gi Alpha1 complexes with two fold parallel symmetry; one of the complexes was deleted for clarity. The catalytic activity of RGS4 and family members is to stabilize the switch regions; in direct contrast with other family of small GTPase activating proteins (such as p120 GAP which accelerated p21ras GTP hydrolysis) which provide sidechain residues to the active site. In Switch 1, notice the central role of T182 of Gi Alpha 1. This residue is conserved in all but the Gs and G12 classes of small GTPases. T182 interacts with seven invariant conserved residues from RGS 4: S85, E87, N88, L159, D163, S164, R167. RGS4 does not associate with T182 suggesting that this residue may contribute to the specificity of RGS4 GiAlpha1 binding. See the reference texts for more details. Shrinivasa, et al. (JBC vol. 273 Jan 16, 1998 p. 1529) reported that the mechanism of RGS4 dependent GTPase activity is through RGS4 stabilization of the "switch" regions. (see kinemage 4 for structure of active site). The mechanism by which RGS4 improves the GTPase activity could occur in two different ways. Since increased GTPase activity is dependent on RGS4/ Gi Alpha 1 binding, either RGS4 could act through the switch regions, or alternatively it could use its sidechains to alter the substrate in some way. It appears that the activity of RGS 4 is mediated through its binding to the switch regions. (kinemage 1) In short, RGS 4 is thought to stabilize the "switches" thus providing a rigid environment for the transformation state of the GDP molecule. (See texts for more complete argument). This has been tested biochemically where substitution of N128 by site directed mutagenesis did not hinder activity, suggesting stabilization of the switch interactions. (JCB vol 273, March 20, 1998 pg 6731.) Another study also states that the binding of RGS4 to Gi Alpha 1 is the important step (JBC vol 272, Jan. 16 1998. pg. 1529.)
