@text Kinemage 1 This kinemage shows the Fab fragment of the indium chelate antibody. It is a murine monoclonal antibody of the IgG 1 type. The monovalent antibody is shown bound to the indium (III)-4-[N'-(2-hydroxyethyl)thioureido]-L-benzyl-EDTA complex.. The Fab fragment consists of two subunits corresponding the heavy (subunit 1) and light (subunit 2) chains of the antibody. Notice that the metal chelate complex has a comfortable binding pocket within the antibody. The antibody-hapten interface is mostly water excluding except for three water molecules involved in hydrogen bonding between the hapten and the antibody. These water molecules are thought to fill holes on the antibody surface. Hydrogen bonds, one salt link (ion pair), van der Waals forces and a histidine-indium coordination are responsible for the high affinity and specificity the antibody has for the In-EOTUBE complex. The EOTUBE molecule consists of an EDTA "head" that's involved in binding with the antibody itself and water molecules at the antibody-hapten interface. It also has a thiourea-ethanol "tail" that has very little interaction with the antibody. Kinemage 2 In this kinemage, subunit 1 of the antibody is seen to indirectly interact with the In-EOTUBE complex through an oxygen atom of tyrosine 34, a nitrogen atom of glycine 52 and a nitrogen atom of asparagine 36. These atoms hydrogen bond to water molecule #1(yellow), which then hydrogen bonds with oxygen 4 of the hapten head. Water molecules 2 and 3 (yellow) are also shown. Notice that the heavy chain interacts with only one side of the hapten. Addition of the light chain (as seen in kinemage 1) forms a snug binding pocket for the hapten. Kinemage 3 In this kinemage, subunit two is shown the interact with the metal chelate complex through an oxygen atom of thr 33 (yellow), three nitrogen atoms of arg 100 (yellow) and a nitrogen atom of histidine 99 (yellow). Two of arg 100's N atoms bond directly to the hapten, while the third N atom hydrogen bonds with water #2 (purple) which subsequently hydrogen bonds with oxygen 1 of the hapten. Histidine 99 is the only amino acid that directly coordinates the indium atom. This is unusual because EDTA usually coordinates metals seven-fold. Kinemage 4 The indium atom is coordinated by two nitrogen and four oxygen atoms from EOTBUE, as well as a nitrogen atom from histidine 99 of the antibody. Coordination by this amino acid is very important because mutation of histidine 99 results in a 20-fold lower affinity of the antibody for the hapten.The hapten has an EDTA head that fits tightly into the antibody binding pocket. The seven bonds coordinating the indium atom are shown. The result of these interactions is no net charge surrounding the indium atom. The other hydrogen bonds come from three water molecules buried at the antibody-hapten interface to the hapten itself. The nature of the hapten also seems to be important in the specificity of the antibody. The addition of the benzyl substituent to one of EDTA's ethylene carbons results in a 24-fold higher affinity of the antibody for the hapten over EDTA alone.
