@text Hello my name is Josh Rausch, and welcome to my kinemage for alpha-hemolysin. This protein was originally crystalized by L. Song et al. (as sited in the reference below) from the bacteria Staph. aureus. Alpha-hemolysin is a toxin which is released from the bacterium as individual monomer subunits which later assemble into their home-heptameric form upon the membranes of erythrocytes. Alpha-hemolysin forms a pore through the membrane in it's final conformation. This pore disrupts the osmotic balance of the cell by allowing ions and just about anything under 10 kD to freely traverse the membrane, in effect turning the cell into a leaky sieve. The binding process is thought to occur in several steps. First the individual subunits are excreted from the bacterium. Subunits then bind to erythrocyte membranes via the flexible loop which later forms the beta-barrel portion of each subunit. In the third step of the formation, all seven subunits come together and form a prepore unit with each subunit linked to the next by the N-terminal tails or "latches". Once the prepore has formed the membrane spanning barrel begins to form and at last the complete pore is established. (D. Engelman Science Vol 274 13Dec 1996, 1850-1851) In this kinemage, and the next one, I have prepared several views for you to illustrate the structure of the alpha-hemolysin heptamer. The preponderance of beta-strands in this protein have been represented using green ribbons. (It is recommended that you only try to look at the specifically selected views and for more detailed inquiry about the structure refer to the next kinemage.) View 1 shows the protein in profile. Here you can see the beta-barrel domain of the protein, which is typiclly refered to as the stem, due to the protein's resemblence to a mushroom. (Though the correct term for that is stype.) The cap domain (the periplasmic exposed portion of the protein) is also apparent, but in this view the rim domain is not readily visible and looks like more of the cap. View 2 gives one a nice view down the pore. If you look closely you can note the rim domains of each subunit as they surround the upper portion of the stem. View 3 provides you with a view of the interior of alpha-hemolysin. Note, looking only at subunits 1 through 4, the increased diameter of the pore in the cap region. This view provides you with a clear look at one of the rim domains.
