@text Class I MHC which is present on the surface of almost all cells, behaves like a receptor that has one binding site for peptide fragments( processed antigen). When a cell has been infected by virus or become a tumor cell which presents non-self peptide, CD8+Tc cells ( cytotoxic T lymphocytes) recognize the peptide in association with class I MHC through TCR, and trigger cytotoxic activity to kill the infected cell. Class I MHC contains a large heavy chain associated noncovalently with a smaller b-microglobulin. Heavy chain consists of a1, a2, and a3 domain. The peptide binding cleft is formed by the membrane distal a1 and a2 domain of the class I MHC. CD8 which is expressed as a homodimer or a heterodimer by Tc cells, recognizes and binds class I MHC. CD8 functions not only to increase the avidity of the interaction between TCR and class I MHC complex but plays an additional role in signal transduction also. Here is a crystal structure of the complex between CD8aa homodimer (residues 1-120) and HLA-A2 (residues 1-276 of the heavy chain, 1-99 of b2-microglobulin) associated with peptide epitope of HIV-1 reverse transcriptase (residues 309-317).
