@text Discovery of Heat Shock protein 15 By using global transcription analysis of the open reading frames in E.coli a number of new heat inducible genes were discovered (approximately 26). The yrfH/hslR gene is among these newly discovered heat shock gene, and it was found that upon heat shock this gene has a 43-fold increase in transcription. The product of the yrfH/hslR gene is a 15,496 Da protein. Because of its molecular weight and its increase upon shift in temperature, this protein is called Hsp15. Characterization of Hsp15 As stated previously, the protein is approximately 15kDa. It has a pI of about 9.94 making it a very basic protein. The normal level of Hsp15 in the cell is ~30um, but when the protein is heat induced the concentration increases to ~60um (2-fold increase). This 2-fold increase is similar to what has been seen with other heat shock proteins such as DnaK. Its sequence was found to have matches with both Gram positive and Gram negative bacteria all of which had similar pI's (b/w 9.9-10.3) and molecular weights (15.1-15.3kDa). When a more sensitive search related to sequence was done, Hsp15 was found to have similarties in motif to certain RNA binding proteins i.e. ribosomal protein,S4 and tyrosyl-tRNA synthetase. This motif is thirty-one amino acids in length and normally begins with RLD and ends in NG. After crystallization of Hsp15 and comparison with these other RNA binding proteins, a new RNA binding motif became evident. This motif has been named the alphaL motif because of the alpha helices involved in the binding and an apparent L shaped meander. This motif is not just limited to a few proteins but is present in greater than 500 proteins. Hsp15 is mostly just this alphaL motif. Role of Hsp15 After discovering the similarities to other RNA binding proteins, the actual affinity of Hsp15 for nucleic acids was determined. It was found that Hsp15 could bind dsDNA, ssDNA and ssRNA. It has a KD value of 4-20(M for dsDNA. From these earlier experiments, it was seen that Hsp15 had a greater affinity for single stranded nucleic acids. Because of its homology to RNA binding proteins and its abundance in the cell, DNA was ruled out as a possible specific substrate for Hsp15. It was determined that the specific substrate for Hsp15 is the free 50S ribosomal subunit. More specifically, Hsp15 will bind 50S subunits only if nascent chains are bound to the ribosomal subunit. Hsp15 will not bind the 50S subunit when it is a part of the ribosome. It is proposed that when there is stress to the cell the ribosome may dissociate before completion of translation, and upon dissociation the nascent chain must be released from the 50S subunit if it is going to be recycled. The interaction that Hsp15 has with 50S and nascent chain may help in releasing the chain and recycling the ribosomal subunit back to the 70S ribosome. Reference: Korber,P. et al.
