@text Tyrosine kinase domain of fibroblast growth factor receptor in complex with inhibitor SU4984 by Lixin Qi Protein tyrosine kinase (PTKs) are essential in signal transduction pathways that control cell proliferation and differentiation. Enhanced PTK activity has been implicated in many human cancers. So selective inhibitors of PTK may have therapeutic value. Fibroblast growth factors (FGF) play important roles in embryonic development, angiogenesis, wound healing and malignant transformation and their function is mediated by four transmembrane receptors (FGFR1 through FGFR4) with intrinsic PTK activity. FGFR1K (FGFR1 Tyrosine Kinase Domain) has a two-lobe architecture typical of protein kinases. ATP binds to FGFR1K in the cleft between the two lobes. SU4984, 3-[4-(1-formylpiperazin-4-yl)-benzylidenyl]-2-indolinone is a compound that can inhibit the function of FGFR1. SU4984 binds to the same region as ATP. The oxindole of the inhibitor occupies the same site as the ATP adenine. The oxindole makes two hydrogen bonds to the backbone of FGFR1K. They are N-1 to the carbonyl oxygen of Glu562 and O-2 to the amide nitrogen of Ala564. The hydrophobic residues including: Val492, Ala512, Ile545, Val561, Ala564 and Leu630 are in the cavity to which oxindole binds.
