@text Transcription initiation requires several factors in addition to RNA polymerase to form a multiprotein complex by interacting with the core promoter elements. Most promoters have a sequence called the TATA box, usually located about 25 bp upstream of the startpoint. The TATA box generally has a relatively fixed location with respect to the transcription startpoint and is found in all eukaryotes. It is characterized by an 8 bp consensus sequence (TATAt/aAt/aA) generally consisting entirely of A-T base pairs. Mutations in this region often significantly effect the ability of the TATA box to bind the necessary factors and function appropriately in transcription initiation. Although eukaryotes have three RNA polymerases that transcribe different classes of genes, they share some common factors. TATA-binding protein (TBP) is one of the factors common to all three polymerases and is involved in the first step of initiation complex formation for genes binding TFIID to the promoter. TFIID consists of TBP as well as several TBP-associated factors (TAF's) which are thought to confer the necessary cell and species specificity to the complex. Therefore, TBP must have the ability to interact with a variety of factors and polymerases that are needed for each type of promoter. TBP itself binds the TATA box with high affinity regardless of the RNA polymerase to be utilized for transcription. The following series of kinemages describes the interaction of the 185 amino acid carboxy-terminal domain of yeast TBP with the TATA box of the CYC1 promoter (Y. Kim et al.). The carboxy-terminal 180 amino acids of the TBP protein are conserved as is the 8 bp TATA box it binds. The proteins N-terminus is variable and can interact with a variety of other factors. Other groups have found strikingly similar interactions by studying the heterologous complexes of Arabidopsis and Human TBP with the TATA box of an adenovirus promoter. TBP is a small protein of approximately 30,000 daltons containing a curved ten-stranded antiparallel B-sheet with 4 a-helixes on its upper suface. The result is a saddle-shaped protein consisting of two subdomains which correspond to the two direct repeats in the amino acid sequence. The structure of TBP bound to the TATA box DNA revealed suprising results. The protein is largely unchanged in structure after contact with the DNA; however, the DNA containing the TATA box is bent and untwisted in order to make contact with TBP along its minor groove in an orientation almost parallel to the TBP molecule. This interaction is stabilized by a variety of hydrophilic and hydrophobic contacts. The specific interactions will be discussed further in the captions of the following kinemages.
