@text The GroES/GroEL/ADP crystal structure affords a view of a new mobile loop conformation that may be compared to the mobile loop conformation trapped by lattice contacts in the GroES-only crystals and the conformer of the GroEL-bound mobile loop peptide described by trNOE NMR (Fig). As expected, the conformer in the GroES/GroEL/ADP complex differs markedly from the conformer trapped by lattice contacts. In the trapped conformation, the loop may be described generally as a twisted hairpin with the hydrophobic tripeptide Ile25-Val26-Leu27 packed against the GroES b barrel. However, NMR studies predicted that the hydrophobic tripeptide would be intimately involved in the binding to GroEL. Partial superimposition of the trNOE structure of the GroEL-bound loop peptide suggested that movement of the C-terminal portion of the mobile loop down and away from the b barrel could result in a more reasonable conformation for binding to GroEL. Indeed, the loop conformation observed in the GroES/GroEL/ADP complex could arise from the proposed movement, and each residue of the hydrophobic tripeptide makes contact with GroEL. Despite the congruence of co-crystal and trNOE structures in terms of the backbone topology and the GroEL-binding residues, differences in structural details point to complexity in the GroES/GroEL/nucleotide complexes that is yet to be resolved. In the trNOE structure, Ser21 lies opposite Ile25; Lys20 is opposite Val26; and the hairpin is flat. In the GroES/GroEL/ADP complex, the register of the hairpin is shifted; Lys20 lies opposite Leu27; and the tip of the hairpin is bent over. Interestingly, the loop conformer trapped in the GroES crystal lattice is shifted in the opposite direction, pairing Lys20 with Gly24. Transitions between these conformations must involve the disruption of several inter-strand hydrogen bonds and the concerted flip of at least two backbone dihedral angles. This is obviously not a problem for the unbound mobile loop which is extremely dynamic. However, could this conformational transition impede discharge of GroES from the GroES/GroEL/ADP complex? We suppose that the trNOE conformation corresponds to the GroES/GroEL/ATP complex. NMR studies show that it is the most favored conformation for the mobile loop and that it can be stabilized in the loop peptide by interactions with the apical domains of GroEL/ADP or GroEL/ATP. In the context of GroES, this conformation may only be accessible when the apical domains of GroEL undergo the full ATP-induced rotation. After ATP hydrolysis, the back-rotation could be resisted by the stability of the mobile loop's most favored conformation and optimal interaction with GroEL binding sites. Such a barrier to GroES dissociation is a recipe for cooperativity the conformational transitions of GroEL subunits, which is likely to be critical for efficient binding and discharge of substrates.
