The segment 177-182 is missing because at this stage the density was not adequate for model building. Although the density for one mobile loop eventually turned up, none were evi dent at this time and thus no mobile loops are shown. The mobile loop trapped by crystal contacts and the conformer generated by trNOE NMR are presented in kinemages 5 and 6. Residue numbering is as follows: subunit 1, 101-197; subunit 2, 1101-1197; subunit 3, 2101-2197; etc. Kinemage 1:GroES C-alpha trace. Hold mouse in middle of structure and move upward to twist structure around the x-axis. (Or simply select View 2) Note the domed shape which is similar to the structure observed in Helen Saibil's electron micrographs of the GroEL/GroES complex. Rotate further to see the cavity underneath (View 3). Kinemage 2: The backbone and sidechains of subunits 1,2,7. Turn off sidechains (except those marked "loop ends" and "in Gp31") for a clearer picture. Scan through the views to get a closer look at the subunit interfaces. Note that conserved positions are dispersed evenly over the structure. Lys15 and Lys34 (ye llow sidechains) flank the mobile loop. The distance between their C-alphas is 4.6 Angstroms. Kinemage 3: Superimposed subunits. Seven-fold symmetry was imposed for most of the subunit during refinement; thus, the bulk each subunit is forced to be the same. However, John freed the symmetry in the "roof loops" because it was evident in the density that the loops were not all the sa me. Note that the roof loop of subunit 2 is significantly different from the rest. (Click on the subunit with the mouse, and the residue number will be returned). Subunit 2 also is the one whose mobile loop is visible, presumably due to crystal contact s with a neighboring GroES molecule. Kinemage 4: Superimposed subunits 1 and 2 with hydrogen bonds. Subunit 2 (yellow) is in the "odd" conformation. coordinates from file: ES2POST.PDB Kinemage 5 One GroES mobile loop trapped by crystal contacts. Highlight subunit 2 by clicking on "su 2".
